2023年8月15日,维克森林大学林慧观团队(陈庭金为第一作者)在Cell Metabolism在线发表题为“NSUN2 is a glucose sensor suppressing cGAS/STING to maintain tumorigenesis and immunotherapy resistance”的研究论文,该研究表明葡萄糖是一个辅助因子结合到甲基转移酶NSUN2的氨基酸1-28,促进NSUN2的寡聚和激活。NSUN2激活维持全球m5 C RNA甲基化,包括TREX2,并稳定TREX2以限制胞内dsDNA积累和cGAS/STING激活,促进肿瘤发生和抗PD-L1免疫治疗耐药性。[查看]

通过基因工程技术将T细胞激活,并装上定位导航到肿瘤的装置-肿瘤嵌合抗原受体(Chimeric Antigen Receptor, CAR),可以将T细胞这个普通“武器”改造成“超级武器”,即为CAR-T细胞。CAR-T是治疗B细胞恶性肿瘤的有效疗法,但在实体瘤中疗效有限。这些限制性的因素包括:肿瘤部位T细胞的迁移和浸润不足、抑制信号的持续存在、肿瘤限制性抗原的缺乏。此外,CAR-T疗法的治疗潜力也取决于CAR-T与肿瘤细胞之间形成裂解免疫突触(lytic immune synapse, IS)的形成。[查看]
